Abstract Apolipoprotein A-I Milano (Apo ) is a natural mutant of Apolipoprotein. It currently the only protein that can clear arterial wall thrombus deposits and promptly alleviate acute myocardial ischemia. Apo considered as most promising therapeutic for treating atherosclerotic diseases without obvious toxic or side effects. However, current biopharmaceutical platforms are not efficient developing . The objectives this research were to express using genetic transformation ability N. tabacum method clone coding sequence into plant binary expression vector pCHF3 with Flag/His6/GFP tag. constructed plasmid was transformed by modified agrobacterium-mediated method, transformants selected under antibiotic stress. PCR, RT-qPCR, western blot co-localization analysis used further verify resistant stable transient established, pure product obtained through A/G agarose. results showed expressed in yield 0.05 mg/g leaf weight purity 90.58% ± 1.65. subjected amino acid sequencing.
